A fuel tanker exploded shortly after midnight at Sachangwan along the Salgaa-Nakuru stretch of the Nakuru-Eldoret Highway, leaving at least two people injured.
The incident occurred in the early hours of Tuesday, prompting a multi-agency emergency response to contain the resulting fire.
Emergency teams attended to the two casualties as firefighters and other agencies worked to put out the blaze. Police said the fire had been contained by Tuesday morning.
The cause of the explosion was not immediately established, with experts expected to investigate the circumstances surrounding the incident.
Police said the highway had been closed at Kibunja and the Soilo interchange as a safety precaution.
Motorists using the Nakuru-Eldoret highway were advised to use alternative routes as emergency teams continued operations at the scene.
Several agencies responded to the incident, with police urging motorists and other road users to exercise caution around the affected section.
Rift Valley Regional Police Commander Samuel Ndanyi said they are investigating the incident. He said emergency teams were checking if there were more casualties at the site.
The incident rekindled memories of a similar tragedy in the same area in 2009 which left more than 100 people dead.
